Normally this should work. The MIDI port assignment should stay intact.
I say "should", because doing so is some work, as when you add a
MIDI
interface, MIDI port enumeration changes. So it's a good idea to check
if the MIDi in and out ports of the US-428 still are "US-428 Control
Port".

This is just a side effect that control surface plug-ins use the same
file format as SoundDiver Modules, and Unitor8 Control is a
stripped-down SoundDiver. Therefore the Finder thinks that the US-428
plug-in can be used by Unitor8 Control.
